This article is a brief tutorial on how to use an optical fiber spectrometer to measure the optical properties of a tissue such as human skin. The discussion presents a typical experiment and the least-squares fitting algorithm for specifying the blood content, mixed arterio-venious hemoglobin oxygen saturation, bilirubin and scattering properties of the tissue.
